Unable to open design file/ duplicate groups in design file

I am attempting to run the amova within version 1.45.3; however, yesterday I continuously received the error “duplicate names in your design file.”

Upon attempting this today, I am receiving the following error:

Unable to open ManVCube.design sets=all. Trying default /Volumes/…/ManVCube.design sets=all.
Unable to open /Volumes/…/ManVCube.design sets=all. Trying mothur’s executable location /Volumes/…/ManVCube.design sets=all.
Unable to open /Volumes/…/ManVCube.design sets=all. Trying mothur’s tools location ManVCube.design sets=all.
Unable to open ManVCube.design sets=all.
Unable to open ManVCube.design sets=all
[ERROR]: did not complete amova.

I tested this with a smaller set and it worked without a problem. The design file was created in excel as a tab delimited, but re-saved as a simple document .design file. Any recommendations would be greatly appreciated.

when following a thread regarding the design file error from last year, the command went through - however, it is now only doing a single comparison in stead of my 8+groups. Could this result as a glitch or would this relate to my design file? The amova goes through using the exact same command using mothur version 1.39.5.

Can you post the command you are trying as well as a screenshot of the error message? From the looks of things, you might be missing a comma between /Volumes/…/ManVCube.design and sets=all
