Hi,

I have a question about mock community. I saw mock community in 454 SOP.
Did the mocke community need me to choice some known species and get them sequences?

Can you give some detail about how to make mock community?

Correct - you need to sequence the mock community in parallel with your actual samples. The mock community can be any collection of DNA that you process in parallel (from PCR through make.shared). So you have to know the “true” sequence of those fragments. You might consider using old clone libraries that have already been sequenced or strains you have laying around the lab and have sequence data for already.
